Uses of Vinblastine
Vinblastine in the treatment of breast carcinoma, trophoblastic tumors, testicular carcinoma, lung, bladder and kidney carcinoma, neuroblastoma, Hodgkin’s and non-Hodgkin’s lymphomas, Kaposi’s sarcoma, Letterer-siwe mycosphonoid disease and treatment of chronic myeloid rheumatic leukemia .
Mechanism of action of Vinblastine
Vinblastine stops cell division in the metaphase phase.
Pharmacokinetics of Vinblastine
Vinblastine crosses the blood-brain barrier. Its metabolism is hepatic and its excretion is mainly through bile and to a lesser extent by the kidneys.
Side effects of Vinblastine
Cardiovascular: angina, cerebrovascular accidents, abnormal electrocardiogram, hypertension (common), ischemic heart disease, ischemia of the hands and feet, myocardial infarction, Raynaud’s phenomenon
Central nervous system: Decreased deep tendon reflex, depression, dizziness, headache, nausea (common), metallic taste in the mouth, neurotoxicity (length: more than 24 hours), limb drowsiness, peripheral neuritis, seizures, tumor pain (common)
Skin: Hair loss (common), dermatitis, blisters, skin sensitivity to light (rare), skin rash
Endocrinology and Metabolism: Elevated Blood Uric Acid, Inadequate Urinary Hormone Discharge Syndrome (SIADH)
Gastrointestinal: Abdominal pain, anorexia, constipation (common), diarrhea, enterocolitis (bleeding), gastrointestinal bleeding, intestinal obstruction, nausea (mild), ileal paralysis, inflammation of the mouth, toxic megacolon, vomiting (mild)
Genitourinary: Azoospermia (lack of sperm), urinary retention
Hematology and oncology: anemia, bone marrow suppression (common), granulocytopenia (common, remission 5-10 days, improvement 7-14 days, dose limiter), uremic hemolytic syndrome, leukopenia (common, remission 5-10 days, improvement 14- 7 days, dose limiter), rectal bleeding, thrombocytopenia (improvement within a few days), thrombocytopenic thrombotic purpura (TTP)
Local: Local stimulation
Nervous, Musculoskeletal: Jaw Pain (Common), Muscle Pain, Bone Pain (Common), Weakness
Ocular: Nystagmus (involuntary eye movement)
Ear: hearing loss, deafness, vestibular disorder
Respiratory: bronchospasm, shortness of breath, pharyngitis
